Abstract

Abstract Background and objectives: Herpes simplex virus type two (HSV-2) infections are mostly considered as a sexually transmitted disease. It causes genital herpes, newborn herpes non-pyrogenic meningitis and may act as an underlying factor for HIV infection. This study was conducted to determine the sero-epidemiologic prevalence of herpes simplex virus type two in cases referred to ten clinical laboratories of Gorgan. Materials and methods: This cross- sectional study was performed on blood Samples of 406 randomly selected Clients. These samples were analyzed for Anti HSV-2 using Specific enzyme-Linked immunoassays (ELIZA) Results: Four point nine percent of all subjects have anti HSV-2. There is Significant difference between sero-positive HSV-2 and factors such as age and Marital Status (P<0.05). Conclusion: HIV is progressively increased and HSV-2 infection is one of the major risk factors for HIV therefore, decreasing the prevalence of HSV-2 infections may Leads to the reduction of HIV prevalence. Abstract

Abstract Backgrounds and objectives: Herpes simplex virus type 1 (HSV- 1) infections are mostly shown as a Herpes disease, but It causes conjunctivitis, genital herpes, encephalitis and newborn herpes. This study was conducted to determine the sero-epidemiologic prevalence of herpes simplex virus type 1 in cases referred to clinical laboratories of Gorgan, Iran. Material and methods: In this cross sectional study, we did random blood sampling on 406 cases referred to the Gorgan city's clinical laboratories. These samples were analyzed for HSV-1 Immunoglobulin G and M antibodies using type- specific enzymelinked Immunoassays (ELISA). Results: Of 406 participants, the HSV-1 seroprevalence is 49% (44.3% and 4.7% for IgG and IgM respectively). There is not significant relationship between seropositive HSV-1 and gender, ethnicity, age and marital status. Conclusion: Sero-epidemiological of HSV-1 in Gorgan is the same of the other places in Iran, but it is higher than European and lower than African countries. It seems that the people’s culture is very important. Therefore it needs to be investigated more. Keywords: HSV-1, Antibody, Gorgan.

Abstract

Abstract Background and objectives: Infection has a Leading role in pregnancy. Cytomegalovirus (CMV), listeria and Toxoplasma are the most common causes of infection in human. Based on the previous researches, about 15-25 percent of being infected during pregnancy leads to some complications such as abortion, fetal death, early labor and etc. This study was designed to determine the seroprevalence of Cytomegalovirus (CMV), Toxoplasma gondii and Listeria moncytogenes among pregnant women in Gorgan, north of Iran (2005-2006). Material and Methods: we conducted this Simple randomized study on 118 unsuccessful pregnant woman and 99 successful ones referred to Deziani hospital in Gorgan. We assayed both IgG and IgM antibodies for CMV and Toxo by Elisa and IFA method for Listeria. In addition, we fill out a Check list and then use SPSS soft ware, chi square to analyze the data. Results: The frequency of IgG for CMV and Toxo is 89.9% and 45.5% in successful pregnant women and 77.1% and 44.1% for unsuccessful pregnant women (P=0.41, P=0.01). IgM frequency for CMV and Toxo is 14.1% and 46.5% in successful women and 30.5 and 21.7% in unsuccessful ones. (P=0.003, P=0.002)Total frequency (IgG, IgM) for Listeria is 7.62% and %3.03 in successful and unsuccessful women, respectively. There is a significant relation between abortion and IgM titer against Toxoplasma in successful and unsuccessful groups. (P=0.003).This relation is true for total antibody titer against Listeria (P=0.003). Conclusion: Because of high titer of antibodies against CMV, Toxo and Listeria in unsuccessful pregnant women, suffering from these agents during pregnancy may result in abortion and fetal death. Hence, we recommend to hold some preventive and educational program and also to assay antibodies against theses agents. Abstract

Abstract Background and objectives: After respiratory infection, Diarrhea is the second cause of mortality. Yersinia enterocolitica is the second important cause of infectious diarrhea in children of some countries. In this study, we evaluated the frequency of Yersinia entocolitica of diarrheal specimens in Gorgan, Iran. Material and Methods: This descriptive cross - sectional Study was carried out on diarrheal stools of 455 patients referred to medical centers and laboratory of Gorgan in 2004-2005. DNA extraction using phenol chloroform was performed for all samples. Using two specific primers (genus-specific16s rRNA and ail- specific species genus of Yersinia enterocolitica), we did PCR sample. Results: Yersinia genome was identified in 12 patients(2.63%) and 11 of them was Yersinia enterocolitica. The frequency infection in of girls (3%) was more than boys (2.4%), and the prevalence in winter (4%) was more them other seasons, and under one- year- group (3.4%) and 1-5 years (3.1%) is more than other age groups. It was not observed significant difference. (P> 0.05). Conclusion: The frequency of Yersinia in cases of diarrhea in Gorgan is similar to most regions of Iran and in children under 5 years is observed more in winter. Abstract

Abstract Background and objectives: Giardiasis is a parasitic infection of small intestine, with a worldwide distribution and the prevalence of Giardia in different parts of the world varies between 1 to 25%. Plants have the vast range of antimicrobial and antifungal activity that can be identified as alternative treatments for bacterial and parasitic pathogens, the same as Giardia. In this study, the methanol extracts of eucalyptus plants, Satureia hortensis and Heracleum glabrescens, on Giardia cysts were studied in vitro. Material and Methods: The cysts were isolated from the feces using a modified Bingham. After counting by Hemusytumetr, they were placed near by 200 mg / ml, 100 mg / ml and 10 mg / ml of the extracts prepared by DMSO for 30 and 60 minutes. Then, the number of dead and live cysts was counted under a microscope. Results: the fatality effect of the extracts in 60 minutes is higher than those of 30 minutes. The methanol extracts of Satureia hortensis, Eucalyptus and Heracleum glabrescens with the dilution of 200 mg/ml in 60 mins have the fatality effect of 84/3%, 63/3% and 44%, respectively. The highest fatality(84.3%) on Giardia cysts is related to Satureia hortensis with the dilution of 200 mg/ml in 60 mins and the Lowest(27%) is related to Heracleum glabrescens with the dilution of 10 mg/ml in 30-minute period. The significant relationship between the plant type and the fatality of methanol extracts is observed. Conclusion: the methanol extracts of Eucalyptus, Heracleum glabrescens and especially Satureia hortensis have anti-parasitic effects in the laboratory conditions. Abstract

Abstract Background and Objective: Cercarial dermatitis (CD) or swimmer’s itch is a severe inflammatory reaction caused by penetration of the skin by avian and herbivorous schistosomes, esp genus Trichobilharzia. Since both intermediate and final host of this parasite exist in North of Iran, we aimed at determining the prevalence of cercarial dermatitis among paddy-field workers in Central areas of Mazandaran Province. Material and Methods: To perform this descriptive-cross sectional study, we refer to bird refuges of migrating water and Paddy-field around man-made ponds of Babol, Sari, Ghaemshahr and Savadkoh districts. The Hand and foot of the farmers were examined clinically for detection of infected people and their signs and symptoms were recorded on a sheet of paper. Results: Based on the results, 77.5 % of the workers suffer from CD. All of them are males and the majority of them are indigenous. Conclusion: High prevalence of Cercarial dermatitis among paddy-field workers led to a health dilemma. Thus, by increasing awareness of farmers, implementing suitable approaches, controlling the disease and sanitizing the contaminated areas can promote health situation. Abstract

Abstract Background and Objective: Bacterial colonization in upper respiratory airways is one of the major risk factors for the development of the ventilator–associated pneumonia (VAP), which is the most common and serious hospital-acquired infection in intensive care unit (ICU). The aim of this study was to determine the frequency of oropharyngeal microorganisms of patients with tracheal tube hospitalized in ICU. Material and Methods: Of 39 patients hospitalized in ICU of panje Azar Hospital, the oropharyngeal cultures were taken after admission. The samples were evaluated for growth of Staphylococcus aureus, Pneumococcus, Enterococcus, Pseudomonas, and E-coli. Results: The mean age of the patients (21 men, 18 women) was 43.64±15.01. The culture was positive in 28.2% and the most common isolate was Pseudomonas aeruginosa (10.3%). Conclusion: Pseudomonas, which is the main pathogen for ventilator- associated pneumonia, may be a potential threat for the patients hospitalized in intensive care units. Abstract

Abstract Background and objective: The periodic evaluation of antimicrobial activity of different antibiotic is essential because antibiotic sensitivity pattern may also changed during short courses. The aim of this study was to assess the frequency of Multi-drug Resistance (MDR) in Gram negative uropathogens. Material and Methods: This study was conducted on 111 gram negative uropathogens using standard microbiology methods in Gorgan, 2011-2012. Antibiotic susceptibility was investigated by Kirby-Bauer disk diffusion methods (DDM). Results: the most common isolates were klebsiella ( 40.5%) , Enterobacter (26.1%) , pseudomonas (13.5%) , proteus( 6.3%) , acinetobacter (1.8% ) and other gram negative bacteria ( 18.3%) .The highest antibiotic resistance was seen to clindamycin (99.1%), and the most sensitivity to Carbapenems (94.6%).Multi drug resistant was seen in 68.5% of isolates. In inpatients, all of the citrobacter species had resistant to multi drugs simultaneously. Conclusion:a high frequency of multi drug resistant in uropathogens is observed in both inpatients and outpatients. Keywords: Multi Drug Resistant, Gram Negative Bacteria, Urinary Tract Infection

Abstract

Abstract Background and Objective: Along with antibiotics, the use of biological methods to combat bacteria is notably considered. A natural barrier such as amniotic membrane is one of the ways of dealing with bacterial infections. The aim of this study was to determine the antibacterial effect of human amniotic membrane. Materials and Methods: This descriptive study was performed in Dezyani teaching Hospital of Gorgan University of Medical Sciences, Iran. To evaluate the antibacterial activity against Staphylococcus aureus, Pseudomonas aeruginosa and Escherichia coli bacteria, 20 amniotic membranes were obtained from postpartum mothers and examined by repeated dilution, diffusion and extraction techniques. Data were collected by observation method and described by mean and standard deviation. Results: The antibacterial activity was found in 15% of the samples against Staphylococcus Aureus and Pseudomonas aeruginosa, while no antibacterial activity was found against E. coli. Given the 15% positive responses, "Diffusion" and "repeated dilution" techniques were more effective in investigating the antibacterial effect of amniotic membrane. Conclusion: The results show the probability of antimicrobial effect of amniotic membrane tissue and it seems that this property can be affected by many factors. Abstract

Abstract

Background and Objective: Staphylococcus aureus is an important cause of food poisoning. Due to the high consumption of fruit and ice cream in summer, this study aimed to determine the prevalence of S. aureus in traditional ice cream and juice in Gorgan, Iran.

Material and Methods: This cross-sectional study was conducted  on 25 places selling traditional ice cream and juice in the summer  of 2014 in Gorgan, Iran.

Results: Results showed that  the microbial contaminants  in traditional ice cream and  juice  centers  were  56%  and 14.28% of S. aureus, 44% and 33.3%  of  yeast, 28% and 28.57%  of B. cereus, 12% and 4.76%  of  Listeria, 8% and 14.28% of Bacillus subtilis and  4% and 4.76%  of  Bacillus licheniformis ,respectively.

Conclusion: The findings of this study indicated a high  contamination of traditional ice cream and juice with Staphylococcus aureus and various microorganisms.

Abstract

Abstract

        Background and Objective: Normal hemoglobin (Hb) is formed of a heme group and a protein group known as globin. Globin is made of four polypeptide chains and in hemoglobinopathies, the structure of one of these four polypeptide chain becomes abnormal. Cellulose acetate method is a common way to differentiate haemoglobinopathies. Inability to identify the components of Hb low concentrations and incapability to isolate all Hb types are among the disadvantages of this method. The aim of this study was to report the prevalence of hemoglobinopathies in the North of Iran by capillary electrophoresis method.

      Methods: All patients with suspected hemoglobinopathies, referred by physicians for electrophoresis, have been studied in a private center in the city of Gorgan, Iran. The level of HbA2, HbA, HbF and other Hb was recorded.

       Results: Overall, 725 blood samples were analyzed using the capillary method. HbE was reported in 2 patients, HbH was observed in 2 patients and Hb Barts was reported in 3 patients. Using the capillary method, among patients with the SDG area, only 4 of 38 (10.52%) had HbS and the majority of them (89.48%) had HbD.

      Conclusion: HbD is the most common hemoglobinopathy in the North of Iran.

Abstract

Abstract

      Background and objective: Hepatitis B virus (HBV) is a DNA virus with high tendency toward hepatic tissue. There are currently about 3 million HBV-infected people and 350 to 400 million chronic carriers of this virus in the world. X protein plays a role in the over-expression of oncogenes, carcinogenicity of liver cells and overlaps with the basal core promoter of the virus. Mutations at specific nucleotides of this region increase viral replication and liver disease progression. The aim of this study was to investigate the frequency of mutations at nucleotides 1762, 1764 and 1766 of HBV X gene in patients with chronic hepatitis B and hepatitis B-related cirrhosis.

      Methods: In this study, 102 patients including 68 chronic hepatitis patients and 34 patients with hepatitis B-related cirrhosis were enrolled. After DNA extraction, HBV X gene was amplified and sequenced using Semi Nested-PCR. Obtained gene sequences were compared with the standard sequence of HBV virus X gene available in the gene bank (Okamoto AB033559). Then, the mutations in the gene X of HBV were identified.

      Results: Comparison of the standard sequence with sequences obtained from patients showed the presence of A1762T / G1764A mutation in 12 chronic (17.64%) and 13 cirrhotic (38.23%) patients. Also, C1766G / G1764T mutations were found in 8.23% of chronic patients and 17.64% of cirrhotic patients.

      Conclusion: A1762T / G1764A mutations in the overlapping region of the basal core promoter with gene X C-terminal may lead to liver disease progression from chronic hepatitis to cirrhosis, by changing the amino acid sequence of the X protein.

Abstract

ABSTRACT

       Background and Objective: Esophageal cancer is the third most common type of cancer in Iran. Studies on water, soil, grains, waste and sediments in Golestan Province have shown that the environmental concentrations of antimony and strontium are increased in areas with high incidence rate for cancer. The carcinogenic effects of strontium have been reported, but carcinogenicity of antimony is still unclear. Since there is not enough information regarding the relationship between the concentration of strontium and antimony and prevalence of esophageal cancer in the province, this study aimed to evaluate serum levels of these elements in patients with esophageal cancer using atomic absorption spectroscopy.

       Methods: This study was performed on 30 patients with esophageal squamous cell carcinoma and 30 healthy matched controls. After sampling, concentrations of strontium and antimony were measured using atomic absorption spectroscopy.

       Results: Half of the esophageal cancer patients and control participants were male. The mean age of patients and controls was 66 ± 13 and 64 ± 8 years, respectively. The mean serum concentration of strontium in patients and controls was 24.18 ± 1.62 and 24.08 ± 1.46 μg/L, respectively. The mean serum concentration of antimony in patients and controls was 18.61 ± 1.48 and 18.98 ± 1.93 μg/L, respectively. No statistically significant difference was observed between the serum concentrations of the two elements in patients and controls.

        Conclusion: There is no difference between serum levels of strontium and antimony in cancer patients and healthy controls, which could be due to tissue accumulation.

Abstract

ABSTRACT

        Background and Objectives: sperm motility mainly depends on aerobic glycolysis, and lactate dehydrogenase (LDH) is a key enzyme in the last step of this process. On the other hand, fructose is considered as the main energy source for sperm motility. Therefore, this study aimed to evaluate the LDH activity and fructose levels and their correlation with sperm motion parameters in normospermic and asthenozospermic males.

       Methods: In this case-control study, 96 normospermic and 96 asthenozospermic males were enrolled.  The semen samples were analyzed by computer assisted semen analysis to identify sperm motion parameters. Seminal plasma fructose levels were measured using Seliwanoff's test. LDH activity in the samples was evaluated using commercially available kits. The obtained data were analyzed by SPSS software (version 16).

       Results: The levels of LDH activity and fructose concentration were not different between the two groups. There was no correlation found between LDH activity and fructose concentration or sperm motility parameters. However, LDH was significantly correlated with sperm concentration in both groups.

      Conclusion: Although LDH activity and fructose levels seem to be influenced by sperm motility, other factors such as sperm concentration and total sperm count can also affect their seminal plasma levels. Thus, when the sperm concentrations are equal in study groups, the level of these factors are expected to be equal.

Abstract

Background and Objective: The cell division cycle 25 (CDC25)is a familyof highly conserved dual-specificity phosphatases that activate cyclin-dependent kinase complexes. These complexes are the main cell cycle regulators. Mammalian cells ,exposure to DNA damaging radiations such as ionizing radiation and ultraviolet light, prevent cell cycle progression by activation of checkpoint pathways and lead to cell death.

      Methods: In this study, mice were exposed to different doses of ionizing radiation. Their total cellular protein was extracted from the bone marrow. After determining and matching the protein concentrations, CDC25A phosphatase levels were measured by western blotting.

        Results: The results showed that exposure to different doses of ionizing radiation in vivo significantly increased the expression of CDC25A compared to control group (P <0.05).

        Conclusion: Exposure to ionizing radiation increases the expression of CDC25A phosphatase, which increases the possibility of tumorigenesis in that area by increasing bone marrow cell proliferation.

Abstract

ABSTRACT
       Background and Objectives: Cellulose is a major component of plant biomass, which can be converted into biofuels and valuable chemicals. The key step in utilization of this organic material is its hydrolysis into soluble sugars. This study evaluated cellulase production by Trichoderma harzianum under different pH values, temperatures and incubation periods with the aim to increase enzyme production and decrease its costs.
Methods: The amount of protein production and the hydrolytic activity of cellulase enzymes including exoglucanase, endoglucanase and β-glucosidase produced by T. harzianum were evaluated using various substrates such as avicel, carboxymethyl cellulose, cellobiose, Whatman grade 1 filter paper under different pH values (4, 4.8, 5.5 and 6.5), temperatures (25, 28 and 34 °C) and incubation times (48, 72, 96 and 120 h).
Results: The optimum condition for cellulase production by T. harzianum is 120 hours of incubation at 25 °C and pH of 6.5.
Conclusion: T. harzianum can be used for the production of all three classes of cellulase. This fungus is suitable for the efficient production of cellulolytic enzymes and reducing the cost of consumables.

Abstract

ABSTRACT
         Background and Objectives: Exposure to ionizing radiation in modern societies is inevitable and can cause a variety of adverse health effects such as cancer and birth defects. Therefore, a reliable, repeatable and sensitive method is required for evaluation of radiation exposure. The aim of this study was to determine the amount of histone H2AX phosphorylation as an indicator of radiation exposure to evaluate the rate of double-strand DNA breakage in irradiated mice.
         Methods: In this study, 15 mice were exposed to different doses of ionizing radiation. After extraction of total protein from bone marrow cells, γH2AX protein was measured by western blotting. Data analysis was performed using ANOVA, Tukey's post hoc test, and the Pearson's correlation test.
         Results: The amount of γH2AX protein in the exposed groups increased significantly compared to the control group (P<0.05).
        Conclusion: The results of this study indicate that exposure to ionizing radiation increases the amount of γH2AX protein in bone marrow cells during the early hours. The protein can be used as a biomarker for monitoring of acute radiation or suspected local radiation exposure.

Taurine is a sulfur-containing amino acid with a wide range of vital biological functions. The aim of this study was to compare plasma levels of taurine between patients with EC and healthy controls.
        Methods: Plasma samples from 36 recently diagnosed cases of EC and 36 healthy adults were analyzed by high-pressure liquid chromatography. Data analysis was done using SPSS (version 16). The t-test was used to evaluate significant differences in the level of taurine between the two groups. P-values less than 0.05 were considered statistically significant.
         Results: Patients with EC had significantly higher levels of plasma taurine compared with the controls (P<0.05).
         Conclusion: Patients with EC have significantly higher levels of plasma taurine compared with healthy individuals. This finding suggests that the measurement of plasma taurine in patients with EC could be useful for the early diagnosis of the disease.

Abstract

ABSTRACT
         Background and Objectives: Capparis spinosa has been used in traditional medicine for various applications including treatment of liver disorders and cancer. We studied the effects of this plant on cell proliferation and morphological characteristics of PLC/PRF/5 liver cancer cell line.
        Methods: After preparing ethanolic extract of the plant, the inhibitory effect of the extract was assessed using MTT assay, and morphological changes were assessed by an inverted microscope.
       Results: C. spinosa ethanolic extract exhibited anti-cancer effects in a concentration-dependent manner. Half-maximal inhibitory concentration of the extract was 1051±4.21 mg/mL. Morphological changes including cell shrinkage, reduction of cell volume and nuclear condensation confirmed the inhibitory effect of C. spinosa on PLC/PRF/5 cells.
        Conclusion: According to the results of this study, extract of C. spinosa seems to be suitable for prevention and treatment of liver cancer. Further studies on animal models could verify the efficiency of the extract against cancer cells.

Abstract

ABSTRACT
         Background and Objectives: Breast cancer is one of the most common cancers among women. Cardiovascular disease is one of the most important complications of breast cancer. The aim of this study was to investigate the effects of six weeks of swimming on malondialdehyde (MDA) and superoxide dismutase (SOD) levels in the heart tissue of mice with breast cancer.
         Method: Breast cancer was induced in 30 female Balb/c mice (6-8 weeks old, weighing 150-200 g) by tumor transplantation. The tumor site was examined every day for the formation or absence of tumor. After becoming familiar with the laboratory environment and swimming pool, the subjects were randomly divided into three equal groups: 1. healthy control, 2. cancer control, and 3. swimming-cancer. The swimming group performed swimming training 60 minutes/day, five days a week, for six weeks. After the intervention, the levels of SOD and MDA in the heart tissue samples were evaluated by enzyme-linked immunosorbent assay. Data were analyzed in SPSS (version 16) using independent t-test at significance of 0.05.
         Results: The results showed that induction of breast cancer was accompanied by a significant increase in MDA and a significant decrease in SOD in the heart tissue of mice. Participation in the six-week swimming significantly reversed these effects.
         Conclusion: Swimming could have beneficial effects in improving oxidative factors in breast cancer patients.
