Abstract

Abstract Background and objectives: Genomic DNA extraction of bacterial cells is of processes performed normally in most biological laboratories therefore, various methods have been offered, manually and kit, which may be time consuming and costly. In this paper, genomic DNA extraction of Staphylococcus aureus was investigated using some laundry detergent brands available in Iran to achieve a rapid and cost effective method. Material and Methods: five-enzyme Taj brand, three-enzyme Saftlan brand ,and Darya and Pak brands without enzyme were used in the concentrations of 10, 20, 40, 80 mg/L. Afterwards, in order to evaluate the efficiency of extracted DNA in downstream processing, PCR test was performed for femA gene in the genome of Staphylococcus aureus. Results: DNA extraction using different concentrations of the brands show that extracted DNA using 40 mg/L Saftlan and Taj brand powders have the best results according concentration (µg/ml) and purity (A260/A280) parameters. These parameters are 387.5 1.88 (Taj), 254.1 2.80 (Softlan), 396.6 1.95 (Manual) and 423.3 2.2 (Kit), respectively. Afterward, the PCR test results by show that DNA extraction using laundry detergents has no effect on its efficiency in order to be used in downstream processes. Conclusion: These results indicate that the proper concentrations of laundry detergents can be used to extract genomic DNA with similar efficiency to kit and manual extraction methods. Abstract

Abstract Background and Objective: Enterococci are Gram-positive members of human gastrointestinal flora,in Dairy products and environment. they have emerged as important causes of opportunistic nosocomial infections in recent years. In this study we aimed to investigat and compare the efficiency of MALDI-TOFmass spectroscopy method through Biochemical and Molecular methods for detecting Enterococcus faecalis and Enterococcus faecium. Materials and Methods:seventhy five clinical samples were collected for biochemical, molecular and mass spectroscopy investigations. Samples were treated with Esculin hydrolysis, Catalase, Pyrrolidonylaminopeptidase, 6.5% NaCl solution, motility, 0.04% Tellurite, L-Arabinose and Sorbitol. Using specific primesallele specific PCR was used.The samples were then analyzed by MALDI-TOF mass spectroscopy and Biotyper 3 software. Results:Enterococcus faecium andEnterococcus faecaliswere detected in thirty and forty two samples, respectively whereas three samples showed both bacterial infections. Using biochemical analysis, two E.faecium isolates were Arabinose negative and one E. faecalis isolates was Telliurite negative. All sampleswere showed correct bands in PCR results but twoof them didn't show clear bands(on agarose gel). In mass spectroscopy analysis all strains were correctly detected and well defined. Conclusion: According to our results, MALDI-TOF mass spectrometry in comparison with Molecular and Biochemical Methodscould be a reliable and accurate method that can easily and quickly identify and differentiate Enterococcus faecium and Enterococcus faecalisin clinical samples. Abstract

Abstract Background and Objective: The Plasmodium falciparum EBA-175, via Sialic acid dependent glycophorin A, binds to red blood cells and thus plays a critical role in cell invasion. Some part of second allele in its gene encoding in FCR-3 (Section F) and CAMP (Section C) can be found. This study aimed to determine the prevalence of Plasmodium falciparum EBA-175KD alleles in southeastern Iran. Material and Methods: In this cross-sectional study, using polymerase chain reaction Nest (Nested-PCR) with specific primers was used for the two parts of the EBA-175 gene to be proliferated. Ninety–four microscopic positive blood samples from individuals infected by Plasmodium falciparum were obtained from four different locations in southeastern Iran. Results: Of 94 positive samples, 88 were antigen EBA-175KD. Genotype CAMP (714 bp) and FCR-3 (to 795 bp), respectively, in 31 (32.97 %) and 49 (52.12 %) were found. Eight samples have both FCR-3 and CAMP. Conclusion: Both of EBA-175KD dimorphic genes were found. The frequency of FCR-3 allele was higher in the South East of Iran. Thus, this pattern can be considered in making Plasmodium falciparum vaccines for this area. Key words: Plasmodium Falciparum Erythrocyte Binding Antigen-175 South-East of Iran

Abstract

Abstract Background and Objective: Stenotrphomonas maltophilia is an opportunistic nosocomial pathogen with high mortality in immunocompromised cases. The aim of this study was to isolate and identify Stenotrphomonas maltophilia in the hospitals’ environment and wards. Material and Methods: In this cross-sectional study, a total of 1108 samples were collected from environment of two hospitals during 12 months. Identification of isolates was performed using biochemical, phenotypic (intrinsic resistance to carbapenems) and molecular methods (amplification of 23S rRNA gene). Results: Of the studied samples, 186 (16.78%) nonfermentative gram negative bacilli (NFGNB) were identified. Amongst NFGNB, 18 (1.62%) isolates were identified as S. maltophiliaby using biochemical tests. Of 18 biochemically identified isolates, 15 (83.3%) were confirmed via PCR. Sinks (40%) and men surgery ward ( 33.3 %( were the most contaminated sites and wards of hospitals, respectively. Conclusion: S. maltophilia is repeatedly isolated from sink which shows that the moist hospital environments need to be considered as a source for dissemination of bacteria. Abstract

Abstract Background and Objective: Bacterial colonization in upper respiratory airways is one of the major risk factors for the development of the ventilator–associated pneumonia (VAP), which is the most common and serious hospital-acquired infection in intensive care unit (ICU). The aim of this study was to determine the frequency of oropharyngeal microorganisms of patients with tracheal tube hospitalized in ICU. Material and Methods: Of 39 patients hospitalized in ICU of panje Azar Hospital, the oropharyngeal cultures were taken after admission. The samples were evaluated for growth of Staphylococcus aureus, Pneumococcus, Enterococcus, Pseudomonas, and E-coli. Results: The mean age of the patients (21 men, 18 women) was 43.64±15.01. The culture was positive in 28.2% and the most common isolate was Pseudomonas aeruginosa (10.3%). Conclusion: Pseudomonas, which is the main pathogen for ventilator- associated pneumonia, may be a potential threat for the patients hospitalized in intensive care units. Abstract

Abstract Background and Objective: The Outbreak of human parasitic diseases associated with the consumption of raw vegetables often occurs in both developing and developed countries. This study aimed to evaluate parasitic contamination of edible vegetables in Golestan Province. Material and Methods: This cross-sectional study was carried out in the cities of Golestan Province for six months, 2012. The samples (N = 100) were randomly chosen among different vegetables (parsley, lettuce, radish, and cress), and examined for the presence of helminthic and protozoan parasitic contaminations following washing, centrifuging and sedimentation. Results: Thirty-seven (37%) were found to have parasitic contamination, and of these 30 (81.1%) and 7 (18.9%) were helminths and protozoa, respectively. The highest rate of contamination was detected in parsley (37.9%), and the lowest in radish (12.0%). Moreover, free living larva with 58.6% and Hymenolepis nana ova with 5.1% were the highest and lowest contaminated rates, respectively. Conclusion: Based on our results, consumed vegetables in Golestan Province is considered as a potential risk for some human parasitic infections. Abstract

Abstract Background and Objective: One of the main causes of increased mortality in cancer patients is bacteremia. On the other hand, antibiotic resistance is the major cause of treatment failure in malignant diseases especially in hematological malignancies. The aim of this study was to diagnose the bacterial strains isolated from blood specimens of cancer patients and to determine their antibiotic susceptibility. Material and Methods: In this cross-sectional study, 0.5 ml of venous blood was taken from 613 cancer patients especially leukemia, and blood cultures and antibiotic susceptibility tests were performed using standard methods. Using disc diffusion method, antibiotic susceptibility was performed with a wide range of antibiotics. Results: Out of 613 cultured specimens, 153 (25%) were found to be positive including 76.47% of gram negative and 23.53% of gram positive bacteria. The most common isolated bacteria were E. coli, coagulase-negative Staphylococci, Klebsiella, Staphylococcus aureus and Pseudomonas aeroginosa, respectively. Conclusion: It seems that Ceftriaxone is the best choice for the treatment of gram negative caused bacteremia and Gentamicin for bacteremia caused by gram positive agents. Given the high level of resistance to the commonly used antibiotics, it seems reasonable to avoid of early and inappropriate use of antibiotics to prevent the development of drug resistant bacteria. Abstract

Abstract

      Background and Objective: Enterococci are relatively nonvirulent bacteria that rarely cause disease. Antimicrobial treatment of Enterococci is often challenging due to their antibiotic resistance. This study aimed to investigate the antibacterial activity of aqueous extract of garlic against Enterococcal isolates.

    Methods: In this descriptive study, 120 Enterococcus isolates including 70 multidrug-resistant isolates were collected from hospitals of Babol, Iran. Isolates’ susceptibility to different antibiotics and the antibacterial activity of garlic extract were assessed using methods of minimum inhibitory concentration (MIC) measurement. The experiments were performed according to the Clinical & Laboratory Standards Institute guidelines, using Tryptic soy broth medium and disc diffusion method.

      Results: Among the 120 Enterococcal isolates, 95 (79.2%) and 25 isolates (20.8%) were E. faecalis and E. faecium, respectively. Of the all Enterococcal isolates, the highest resistance was to erythromycin (95.8%), tetracycline (88.3%) and ampicillin (65.8%). While, the minimal level of resistance was to chloramphenicol (6.8%), vancomycin (20%) and ciprofloxacin (25%). Also, 53.3% of Enterococcal isolates showed simultaneous resistance to at least three antibiotics (tetracycline, erythromycin and ampicillin). Such resistance in E. faecium isolates was higher compared to E.faecalis (68% vs. 55.7%). The range of antibacterial activity of garlic extract against isolated Enterococci was determined by growth inhibition zone of 16.8 ± 1.8 mm and MIC of between 4 to 32 mg/ml.

      Conclusion: This study indicates the clear anti-enterococcal effect of aqueous extract of garlic and confirms the use of garlic in treatments by medicinal plants.

Abstract

ABSTRACT

         Background and Objective: Hospital clothing and textiles have a great role in providing medical services to patients. Hospital services would not be possible without using these textiles. The aim of this study was to prepare non-woven polyester for use in medical textiles.

         Methods: In this study, most important expected properties of hospital clothing were created on polyester fabrics via various stages of finishing using chemicals such as calcium chloride, sodium alginate, chitosan and nanoparticles of silver nitrate and aluminum oxide. Antibacterial (activity against Staphylococcus aureus), deodorizing and wicking properties of samples were evaluated using nano-biomaterials via standard tests.

          Results: The water absorption rate of nonwoven samples that were finished with chitosan significantly improved. The results also showed favorable level of antimicrobial activity caused by silver nitrate and chitosan. In addition, deodorizing activity was observed in all samples. However, silver nitrate-finished samples had higher deodorizing activity. The capillary action of all samples reduced after completion of all finishing procedures.

          Conclusion: The results show that the use of chitosan and alginate along with nanoparticles of silver nitrate and aluminum oxide can be effective in achieving the desired properties of medical textiles.

Abstract

ABSTRACT
         Background and Objectives: The present study aimed to investigate effects of eight weeks of aquatic exercise and resistance training on plasma neurotrophin-4 (NT-4) levels and NT-4 expression in peripheral blood mononuclear cells (PBMCs) of women with multiple sclerosis (MS).
         Methods: Thirty women with MS were randomly assigned to a resistance-training group, an aquatic exercise group and a control group. After separating plasma and PBMCs from blood samples, level of plasma NT-4 and NT-4 expression was measured after eight weeks of exercise via ELISA and Real-time PCR, respectively.
        Results: The level of plasma NT-4 and NT-4 expression increased significantly following eight weeks of aquatic training and resistance training. 
        Conclusion: Based on the results of the present study, both resistance training and aquatic exercise can increase the level of plasma NT-4 and NT-4 expression in female MS patients. It can be concluded that such trainings could have protective and positive effects on the nervous system of MS patients.

Abstract

ABSTRACT
          Background and Objectives: Oxidative stress is a common mediator in pathogenicity of established cardiovascular risk factors. The purpose of this study was to determine effects of selected aerobic training on some oxidative stress indicators in sedentary elderly women.
          Methods: In this quasi-experimental study, 21 participants (age range: 60-70 years) were selected by convenience sampling and then randomly assigned to an exercise group (n=11) and a control group (n=10). Aerobic exercise was performed at 50-70% of heart rate reserve for 45-60 minutes per session, three sessions a week, for eight weeks. Blood samples were taken at baseline and 24 hours after the last training session. Within-group and between-group differences were assessed using paired sample t-test and independent t-test, respectively. Statistical significance level was set at 0.05.
          Results: Weight, body mass index and body fat percentage decreased significantly after the exercise intervention (P<0.05). We also noted a significant reduction in endothelin-1 levels in both groups (P<0.05). Nitric oxide increased significantly after the eight-week exercise training (P<0.05). Moreover, serum 8-hydroxydeoxyguanosine levels decreased significantly in the exercise group (P<0.05). No significant change was observed in malondialdehyde and urinary 8-hydroxydeoxyguanosine levels in both groups. The result also showed a significant difference in subjects’ weight and in nitric oxide and 8-hydroxydeoxyguanosine levels between the two groups (P<0.05).
          Conclusion: It seems that regular aerobic exercise at moderate intensity can improve antioxidative capacity and reduce the risk of cardiovascular disease in elderly women.

Abstract

ABSTRACT
          Background and objectives: Visfatin is a newly characterized adipokine, which is highly expressed in visceral adipose tissue. The aim of this study was to evaluate effects of aerobic exercise training on visfatin, homocystein, C-reactive protein and lipid profile in sedentary men.
          Methods: In this quasi-experimental study, 27 sedentary men were selected by convenience sampling method. The subjects were divided into a control group (n=13) and an aerobic training group (n=14). The subjects in the experimental group performed 60 minutes of aerobic exercise at 75-85% of maximum heart rate, three sessions per week for eight weeks. Data were analyzed in SPSS 16.0 (SPSS Inc., Chicago, IL, USA) using paired and independent sample t-test for comparison of means within and between groups, respectively. A P-value of less than 0.05 was considered statistically significant.
          Results: The eight-week aerobic exercise training significantly reduced body weight (P=0.02), body mass index (P=0.01) and low-density lipoprotein-cholesterol (P=0.03) levels. It also caused a significant increase in the high-density-lipoprotein-cholesterol (P=0.00) and a significant decrease in visfatin levels (P=0.005). Serum homocysteine and C-reactive protein levels decreased by the end of the training program, but these changes were not statistically significant (P>0.05).
          Conclusion: The eight-week aerobic exercise program could effectively improve lipid profile and visfatin levels. Therefore, this type of exercise could be recommended for prevention of cardiovascular disease and improvement of its risk factors.

Abstract

Background and objectives: Medicinal plants have long been considered as one of the most important pillars of traditional medicine. Existing challenges in the treatment of diseases, particularly infectious diseases, are major drivers for herbal medicine studies. Tribulus terrestris has been widely used in traditional medicine to treat various diseases. This study aimed to investigate in vitro antibacterial effect of the aqueous extract of T. terrestris on several oral bacteria.

Methods: In this experimental study, after preparing the aqueous extract of T. terrestris, minimum inhibitory and bactericidal concentrations (MIC and MBC) of the extract were determined against standard strains of Streptococcus mutans, Staphylococcus aureus, Klebsiella pneumoniae and Streptococcus pyogenes using the broth microdilution method. The experiments were repeated three times and the results were analyzed with SPSS 22 using the one-way analysis of variance (ANOVA) and LSD statistical tests with the significance level set at 0.05.

Results: The aqueous extract of T. terrestris had the highest inhibitory effect on S. pyogenes and S. mutans, and the difference between the MIC and MBC values was significant (P <0.05). However, no such effect was observed against S. aureus and K. pneumonia at concentrations below 50 mg/ml when compared to ampicillin and chlorhexidine.
Conclusion: The aqueous extract of T. terrestris has significant antibacterial effects against S. pyogenes and S. mutans. Therefore, it can be incorporated into topical formulations such as toothpaste and mouthwash products after further in vivo and toxicity experiments.

Abstract

Background: Research has shown that the extract of some plants has an effective protective effect on liver cells against toxins and oxidants. In this research, the effect of hydroalcoholic extract of ginger against hepatotoxicity caused by doxorubicin (Dox) in adult male rats was investigated.
Methods: In this experimental study, 63 adult male rats were divided into 7 groups (n = 9 per group). The experimental treatments included the control and placebo groups without the use of medicines, experimental group I that received Dox at a dose of 20 mg/kg, experimental groups II and III with 300 and 600 mg/kg of ginger hydroalcoholic extract, and experimental groups IV and V that received 20 mg/kg Dox + 300 and 600 mg/kg of ginger hydroalcoholic extract, respectively. After the end of the test period, serum levels of total bilirubin and direct bilirubin were measured. In addition, the histological changes in the liver were examined after hematoxylin and eosin (H&E) staining.
Results: Serum levels of total bilirubin and direct bilirubin in the Dox group showed a significant increase compared to the control group. In contrast, serum levels of total bilirubin and direct bilirubin in Dox + ZIN 600 and Dox + ZIN 300  had a significant reduction compared to the Dox group (P < 0.05). Ginger extract prevented apoptosis and Dox-induced liver tissue damage in dose-dependent designs.
Conclusion: The hydroalcoholic extract of ginger improves the changes of serum bilirubin and liver tissue after receiving Dox due to its antioxidant compounds.
