Abstract

Abstract Background and objectives: Air ionization (AI) may reduce the microbial content of the air 'and has bactericidal effects on some bacterial Strains, which is controversial. We conducted this study to evaluate the efficacy of Air ionizer in reducing the microbial content of air. Material and Methods: This experimental Study was carried out in Sadi Hospital of Isfahan. Two air ionizer were placed in two boxes. and the third box (Control) was empty. At the beginning of experiment and every 30 minutes up to three hours, we did air sampling in all groups. After taking the samples of every box, we incubated the samples and then performed colony counts and bacteriologic studies. Results: In all thirty-minute intervals, Colony Counts in all groups were lower than control group (P<0.05). 'Coagulase negative staphylococcus (CNS) was the most common bacteria isolated followed by Bacillus spp, Acinetobacter and Escherichia Coli, in control group, no colonization of Acinetobacter and E.coli is found in Air ionizer groups. Conclusion:Our findings qualitatively indicate that air ionization can reduce the microbial content of the air. Regarding the type of microbial air pollution and the amount of air cleaning needed, this method can be used solely or in combination with other air cleaning methods. Abstract

Abstract Background and Objective: Diabetes mellitus is one of complications that thalassemia major patients face with. Hence, blood glucose monitoring is of vital importance to these patients. Because of high level of fetal hemoglobin in these patients, the measurement of hemoglobin A1c is not reliable and should be displaced by fructosamine test. Material and Methods: The current descriptive study was carried out on 33 beta-thalassemia major patients afflicted with diabetes mellitus (21 female and 12 male cases). Blood glucose level, fructosamine, hemoglobin A1c, serum ferritin and fetal hemoglobin were measured. Results: Blood glucose levels are 204±103 mg/dL and 221±101 mg/dL (p=0.63) fetal hemoglobin levels are 9%±7% and 13%±9% (p=0.22) serum ferritin levels are 1744±1534 ng/mL and 3253±1773 ng/mL (p=0.96) in female and male patients, respectively. The level of fructosamine (42±124 mmol/L) and glycosylated hemoglobin (8.9%±1.8%) are correlated significantly (r=0.69, p<0.01). Both Hemoglobin A1c (r=0.75, p<0.01) and fructosamine (r=0.54, p<0/01) show a significant correlation with blood glucose level. Conclusion: In diabetic thalassemia major patients with frequent blood transfusion, the level of fructosamine and glycosylated hemoglobin are related significantly, therefore they can be used alternatively. Abstract

Abstract Background and Objective: The Plasmodium falciparum EBA-175, via Sialic acid dependent glycophorin A, binds to red blood cells and thus plays a critical role in cell invasion. Some part of second allele in its gene encoding in FCR-3 (Section F) and CAMP (Section C) can be found. This study aimed to determine the prevalence of Plasmodium falciparum EBA-175KD alleles in southeastern Iran. Material and Methods: In this cross-sectional study, using polymerase chain reaction Nest (Nested-PCR) with specific primers was used for the two parts of the EBA-175 gene to be proliferated. Ninety–four microscopic positive blood samples from individuals infected by Plasmodium falciparum were obtained from four different locations in southeastern Iran. Results: Of 94 positive samples, 88 were antigen EBA-175KD. Genotype CAMP (714 bp) and FCR-3 (to 795 bp), respectively, in 31 (32.97 %) and 49 (52.12 %) were found. Eight samples have both FCR-3 and CAMP. Conclusion: Both of EBA-175KD dimorphic genes were found. The frequency of FCR-3 allele was higher in the South East of Iran. Thus, this pattern can be considered in making Plasmodium falciparum vaccines for this area. Key words: Plasmodium Falciparum Erythrocyte Binding Antigen-175 South-East of Iran

Abstract

Abstract Background and Objective: Dermatophytes are the fungi that have the ability to attack the keratinized tissues such as the skin, hair and nails. Infections caused by these organisms are named dermatophytosis. We aimed to compare Minimum inhibitory concentration (MIC) of Crude extracts of Active Actinomycete Isolates with Terbinafine, Griseofulvin, Ketoconazole and Fluconazole Drugs against Microsporum Canis, Microsporum gypseum and Trichophyton mentagrophytes. Material and Methods: In this experimental study, in order to find MIC by actionmycete, 100 isolates were studied and then crude extracts of the active actinomycete isolates were prepared in sterile conditions. Finally, the crude extracts obtained at different concentrations were used to obtain the MIC of Microsporum Canis, Microsporum gypseum and Trichophyton mentagrophytes. Moreover, various concentrations of the drugs such as terbinafine, griseofulvin, ketoconazole and fluconazole in solvent Dimethyl sulfoxide (DMSO) were prepared and their growth inhibitory effect was evaluated and then compared with the results obtained from the crude extract of active actinomycete isolates. Results: the crude extracts obtained from active Actioiomycetes isolates and the drugs such as terbinafine, griseofulvin, ketoconazole and fluconazole, in a dose-dependent manner, could inhibit the growth of Microsporum Canis, Microsporum gypseum and Trichophyton Mentagrophytes. Conclusion: compared to MIC of Crude extract of active actinomycete isolates, Terbinafine has a significant effect on the growth inhibition in all of the fungal Dermatophytes and then griseofulvin, ketoconazole and fluconazole are in the next rank, respectively. Abstract

Abstract

Background and Objective: Oral candidiasis is a common opportunistic infection in diabetic patients. Candida albicans is colonized on different oral surfaces such as tongue, palate, dental caries and plaques.  This study aimed to determine the prevalence of   Candida albicans in the mouth of   diabetic patient.

Material and Methods: This cross - sectional study was conducted on 120 diabetic patients. After completing the questionnaire, the   saliva samples were obtained and cultured   on sabouraud agar medium containing 0.005% chloramphenicol. For final diagnosis, Germ tube test and Sugar absorption test (with API 120 kit) were used.

Results:  Of 120 patients, 61 (50.8%) were positive for Candida.  Candida infection rate was relatively higher in older patients and in women.  The rate was significant in the patients using dentures (P< 0.001) and   the ones keeping on smoking (P <0.038).  The infection in the patients with type II diabetes was higher than the cases with Type I.

Conclusion: Given the high prevalence of Candidiasis and Candida colonization in the mouth of diabetic patients, it is a necessity to identify and treat these infections in order to prevent further complications.

Abstract

ABSTRACT

        Background and Objective: The current challenge of diabetes mellitus is to prevent its complications. These complications are directly associated with hyperglycemia in diabetics. The HbA1c measurement is essential for long-term glycemic control. Synchronization of HbA1c measurement is important in order to avoid discrepancies between results reported by laboratories. This study aimed to evaluate the accuracy, precision and agreement of five HbA1c measurement methods with HPLC reference method.

       Methods: HbA1c levels of 55 samples were measured using six methods of microcapillary electrophoresis (Sepia), enzymatic method (Pishtaz Teb), immunoturbidometry (Pars Azmoon), boronate affinity (Nycocard), immunofluorescence (ichroma) and Tosoh G8 HPLC.

       Results: The five tested methods showed a good agreement with the HPLC method with correlation coefficient of less than 95%. Regression testing of HPLC method and other methods showed slope of 0.99 (P<0.05) for Sebia, 1.02 (P<0.05) for Pishtaz Teb, 0.79 (P<0.05) for Pars Azmoon, 0.82 (P<0.05) for Nycocard and 0.89 (P<0.05) for ichroma. Average inaccuracy for the Sebia, Pishtaz Teb, Pars Azmoon, Nycocard and ichroma in comparison with the HPLC reference method were -0.09, -0.004, -0.75, -0.79 and -0.78, respectively.

         Conclusion: The Sebia microcapillary method and Pishtaz teb enzymatic method have appropriate accuracy and precision. Therefore, these methods can be used as alternatives to the HPLC method for HbA1c measurement. Other methods such as Pars Azmoon, Nycocard and ichroma have significant shortcomings in terms of accuracy.

Abstract

ABSTRACT

          Background and Objective: The present study aims at investigating the possible effect of 8-week aerobic exercise on undercarboxylated osteocalcin and beta cell function in postmenopausal women.

         Methods: The study included 20 postmenopausal women with mean weight, height and body mass index of 78.94 ± 5.72 kg, age 48.69 ± 3.21 years, 160.37 ± 4.12 cm and 30.72 ± 2.37 kg/m2. The participants were randomly selected and divided into experimental and control groups. Blood samples were taken 48 hours before the experiment and after eight weeks of exercise. Aerobic exercise was performed for eight weeks, three sessions per week with intensity of 65-70% of heart rate. Data analysis for intragroup and intergroup differences was done using paired and independent t-test, respectively. P-values less than 0.05 were considered statistically significant.            

         Results: The level of undercarboxylated osteocalcin in the experimental group decreased significantly compared to control group (P<0.049). The level of beta cell function index in the experimental group increased significantly after the 8-week exercise program compared to  control group (P<0.014).

         Conclusion: Exercise increases the level of undercarboxylated osteocalcin in postmenopausal women that has important consequences, especially for those at risk of developing diabetes.

Abstract

ABSTRACT
         Background and Objectives: Due to adverse effects of pentavalent  antimonial  treatment of visceral leishmaniasis using herbal medicines has received a lot of attention. In this study, we evaluated inhibitory activity of Pelargonium roseum essential oil on growth of Leishmania infantum promastigotes, and compared it with that of Glucantime.
        Methods: Inhibitory effects of different concentrations (1, 2.5, 5, 10, 25, 50 and 100 µL/ml) of P. roseum essential oil on the growth of L. infantum promastigotes were studied by MTT assay after 24 and 48 hours. All the data were analyzed by one-way analysis of variance. The effect of the essential oil was later compared with that of standard dose of Glucantime.
        Results: After 24 hours, different concentrations of the essential oil had no significant inhibitory effect on promastigotes of L. infantum. After 48 hours, all concentrations except for 1µL/ml showed significant anti-leishmanial activity. In addition, the lowest concentration that had significant inhibitory effect on L. infantum promastigote was 5 µL/ml.
       Conclusion: The essential oil of P. roseum has favorable inhibitory activity on the growth of L. infantum promastigotes after 48 hours. This indicates the therapeutic potential of this plant for treatment of leishmaniasis.

Abstract

ABSTRACT
            Background and Objectives: Nowadays, infections with antibiotic-resistant bacteria are among the most important causes of mortality worldwide. This has attracted the attention of researchers to seek suitable alternatives for antibiotics. The venom of many toxic species such as arthropods has antibacterial properties. In this study, we investigated antibacterial effects of crude venom of Latrodectus dahli on Escherichia coli, Staphylococcus aureus, and Bacillus subtilis.
            Methods: Lyophilized crude venom of L. dahli was dissolved in 50 mM Tris-HCl buffer. Protein concentration was determined by the Bradford assay. Then, the bacteria were exposed to different concentrations (31.25-250 ng/mL) of the crude venom. Inhibitory activity of the venom against the bacteria was determined by MTT assay and determining minimum inhibitory concentration (MIC).
            Results: Results of the MTT assay showed that the crude venom significantly inhibited the growth of E. coli (31.25 and 62.5 ng/mL), S. aureus (at 250 ng/mL) and B. subtilis (at 125 and 250 ng/mL). In the MIC experiment, the crude venom significantly inhibited the growth of E. coli (at concentrations of 31.25 and 62.5ng/mL), S. aureus (at concentrations of 31.25-250 ng/mL) and B. subtilis (at concentrations of 31.25-250ng/mL).
            Conclusion: The crude venom of L. dahli and its components showed relatively strong antibacterial effects.

Abstract

ABSTRACT
          Background and objectives: Nosocomial infections caused by antibiotic resistant bacteria is a life threatening health challenge. This study aimed to determine the frequency of antibiotic resistance genes in clinical isolates from hospitals of Zahedan, southeast of Iran.
           Methods: Overall, 818 isolates were collected from different hospital wards. The isolates were identified using conventional microbiological and biochemical tests. Antibiotic susceptibility pattern was assessed by agar disc diffusion method and determination of minimum inhibitory concentration of a number of antibiotics. Multiplex PCR was performed using specific primers for the detection of resistance genes.
           Results: The most common species were Staphylococcus aureus (25%), Klebsiella pneumoniae (22%) and Pseudomonas aeruginosa (14%). The rate of methicillin resistance among S. aureus, S. epidermidis and S. saprophyticus was 60%, 43% and 24%, respectively. In addition, 28.5% of enterococci isolates were vancomycin resistant. Among gram-negative bacteria, 45% of A. baumannii and 24% of P. aeruginosa were identified as ESBL. A high level of resistance to ampicillin (96%), cefotaxime (89%), gentamicin (89%) and sulfamethoxazole-trimethoprime (60%) was observed in K. pneumoniae.
           Conclusion: Our results highlight the urgent need for an eradication program and a surveillance plan for preventing increased emergence of antibiotic resistant bacteria in the study area.
