Abstract
Abstract Background and objectives: Paratuberculosis or John's disease is a chronic infectious disease of ruminants caused by Mycobacterium avium subsp. paratuberculosis (MAP). It results in major economic losses to dairy farm of all over the world and it is the agent causing crohn's disease. The aim of this study was to detect the MAP using PCR in raw-milk samples of cows in shahre-kord. Material and Methods: In this cross–sectional study, 100 raw milk samples of cows were collected from both industrial and semi -industrial farms in shahre-kord. The DNA of all Samples was isolated by MAP, using PCR method. Results: The results Show that only three (3%) Samples were positive for Mycobacterium avium subsp. paratuberculosis. Conclusion: Based on our results, Milk -PCR was useful for detection of MAP in milk samples. Key words: Mycobacterium paratuberculosis, milk, polymerase chain reaction.

Abstract
Abstract
Background and objectives: Genomic DNA extraction of bacterial cells is of processes performed normally in most biological laboratories therefore, various methods have been offered, manually and kit, which may be time consuming and costly. In this paper, genomic DNA extraction of Staphylococcus aureus was investigated using some laundry detergent brands available in Iran to achieve a rapid and cost effective method.
Material and Methods: five-enzyme Taj brand, three-enzyme Saftlan brand ,and Darya and Pak brands without enzyme were used in the concentrations of 10, 20, 40, 80 mg/L. Afterwards, in order to evaluate the efficiency of extracted DNA in downstream processing, PCR test was performed for femA gene in the genome of Staphylococcus aureus.
Results: DNA extraction using different concentrations of the brands show that extracted DNA using 40 mg/L Saftlan and Taj brand powders have the best results according concentration (µg/ml) and purity (A260/A280) parameters. These parameters are 387.5 1.88 (Taj), 254.1 2.80 (Softlan), 396.6 1.95 (Manual) and 423.3 2.2 (Kit), respectively. Afterward, the PCR test results by show that DNA extraction using laundry detergents has no effect on its efficiency in order to be used in downstream processes.
Conclusion: These results indicate that the proper concentrations of laundry detergents can be used to extract genomic DNA with similar efficiency to kit and manual extraction methods.

Abstract
Abstract Background and objectives: The increase of ESBL producing E.coli can create a tremendous difficult y for the health system. These isolates leads to rapid transmission of causative genes to other clinically important bacteria and synchronously increased resistant to other antibiotics. This study was carried out to determine the prevalence of this isolate and related genes in Gorgan, North of Iran. Material and Methods: This study was conducted on 218 isolated E.coli from urinary tract infection of outpatients referring to six medical laboratories in Gorgan, during 2010-11. The resistance to Cefotaxim (Mast Co.) was assessed by Kirby-Bauer disk diffusion method. The confirmatory test for detection of resistant isolates was carried out by double disk method at the presence of Cefotaxim and clavulanic acid. The presence of β lactamase gene of blatem, blactx and blashv in ESBL was assessed by PCR method. Results: of 218, 70 isolates (32.1%) are resistant to Cefotaxim. Sixty-two (88.6%) of them are confirmed as ESBL producing E.coli. β lactamase genes of blactx, blatem and blashv can be seen in 28(45.2%), 26(41.9%) and 6(9.7%) isolates, respectively. Conclusion: the prevalence of ESBL producing E.coli in Gorgan is in the range of country average and blaCTX-M gene is the most common gene. Key words: E.coli, ESBL, bla gene, UTI

Abstract
Abstract
Background and objectives: the increasing use of antibiotics, especially the third generation cephalosporins, is an important factor in the spread of antibiotic resistance in bacteria. The main reason for the development of resistance phenotype such as Extended Spectrum Beta Lactamas (ESBL) is the extensive use of broad-spectrum cephalosporins. In phenotypic survey, the Phenotyping confirmatory test and the minimum inhibitory concentration (MIC) are used. In this study, the prevalence of the isolates resistant to third generation cephalosporin (cefotaxime) was determined based on MIC.
Material and Methods: form September 2010 to September 2011, 75 isolates of Klebsiella pneumoniae were collected from the infections of inpatients and outpatients, referred to state and private laboratories of Gorgan. For all of the Klebsiella pneumoniae strains, MIC determination using E-test (company Liofilcheme-Italy) was performed.
Results: According to the MIC results, 26 samples (34.6%) are resistant to cefotaxime 22 isolates are completely resistant to concentration of 256μg.
Conclusion: Because of the importance of risk of becoming ESBL, further studies are needed to clarify the ESBL in the region.

Abstract
Abstract Background and Objective: Listeria bacteria with worldwide widespread are commonly found in soil, sewage, dust and water. Among which,Listeria monocytogenes can cause a serious food-borne disease. The objective of this study was to investigate the prevalence of Listeria species in ready-to-eat foods. Material and Methods: The samples (n=235) including oloveyh salad (n = 64), Yogurt stew (n= 35), vegetable salad (n=52), macaroni salad (n= 48) and meat salad (n =36) were collected from the restaurants in Shahrekord, Iran. Enrichment and selective media were used to determine the prevalence of Listeria species. . Results: The results showed that 8.5 % of the samples were infected by Listeria spp. The highest were isolated from vegetable salad (17.3%) and the lowest from macaroni salad (4.2%). Listeria monocytogenes was isolated from 7 samples (3.0%). Other isolated species were L.innocua (4.7%) and L.seeligeri (0.9%). Conclusion: It seems that eating ready-to- eat food or raw and undercooked foods has the potential risk of contamination to the Listeria species. Keywords: Listeria, Ready- to- Eat Food, Shahrekord

Abstract
Background and Objective: Biofilm is a complex microbial community embedded in a self-produced extracellular polymeric matrix. We aimed to study the extent of biofilm formation by S. Areas isolates and its relation to some phenotypic and genotypic criteria.
Material and Methods: One hundred-fifty strains of Staphylococcus aureus isolated from Gorgan were studied. Microtiter plate assay method was used for investigation of biofilm formation.The biofilm formation of strains were recorded and its relation to accessory gene regulator (agr) and antibiotic resistance were assessed by X2 test.
Results: Eighty-four isolates (56%) were able to form biofilm. The strength of biofilm formation in agr group I was more than that of other groups. The biofilm formation among S. Areas isolated from the wound and urine (both with 75 %) had the highest capability. Methicillin-resistant isolates had a greater ability to biofilm formation.
Conclusion: Methicillin resistant isolates had a greater ability to biofilm formation. Given the importance and treatment related problems of Methicillin-Resistant Staphylococcus Aureus (MRSA) especially Community Acquired-Methicillin-Resistant Staphylococcus Aureus (CA-MRSA), it is a necessity to control or remove the biofilm formation alongside antibiotic treatment.

Abstract
Abstract Background and Objective: Klebsiella pneumoniae is one of the agents causing nosocomial infection therefore, we decided to report the prevalence of Klebsiella pneumoniae caused infection. Material and methods: The frequency of Klebsiella in culture media samples of Panje Azar hospital was studied in 2011-2012. After determination of the species with biochemical methods and determination of resistance to third generation cephalosporins, the existence of responsible genes for this resistance was investigated using specific primers. The PCR product for CTX-M gene was sequenced. Results: During the study, 70 isolates of Klebsiella were isolated in that 51 (72.8%) related to three months of November, December and January. Except for the one related to November, other ESBL cases belonged to these three months. Based on molecular investigation of ESBL genes, these isolates at least were in 3 types and had a high frequency in Internal, female and Emergency wards. Conclusion: The present report implied a sudden prevalence of Klebsiella pneumoniae that detected and controlled by a correct monitoring. Keyword: Klebsiella Pneumoniae, ESBL, CTX-M

Abstract
Background and Objective: Cryptosporidium parvum is a zoonotic protozoan parasite causing diarrheal cryptosporidiosis. Numerous outbreaks of cryptosporidiosis have been reported worldwide. The transmission via milk, water and raw animal products is one of the important ways. The aim of this study was the identification of hsp70 gene in Cryptosporidium parvum in raw cow’s milk samples.
Material and Methods: In this cross sectional study, 38 raw cow’s milk samples of bulk tank were randomly collected from traditional and semi industrial cattle farms in Isfahan. To identify the protozoa in milk samples, the extracted DNA was evaluated by Nested polymerase chain reaction (PCR).
Results: Based on Nested polymerase chain reaction, 2 samples (5.26%) were infected to Cryptosporidium parvum.
Conclusions: The contamination of milk with Cryptosporidium Parvum is less than that of the other foodstuffs. Thus, it is necessary to reduce food contamination and to have appropriate health education programs.

Abstract

Background and Objective: BabA2 and Hpa genes are involved in adherence of Helicobacter pylori (H.pylori) to gastric mucosal tissue. This study aimed to investigate the frequency of these genes in isolates of H. pylori from gastric biopsies and their relationship with gastritis, peptic ulcer and gastric cancer.
Methods: Gastric biopsy samples were obtained from patients with gastritis, peptic ulcer and gastric cancer. A sample was sent to the laboratory for urease test and histopathology study, and another sample for DNA extraction. The frequency of BabA2 and Hpa genes was investigated using their specific primers by PCR.
Results: Among the 80 analyzed biopsy samples, 51 (63%) were BabA2 positive, and the frequency of this gene in the samples of gastric cancer, gastritis and peptic ulcer was 61.1, 58.3 and 73.3%, respectively. In addition, 57 samples (71%) were Hpa positive, and the frequency of this gene in the samples of gastric cancer, gastritis and peptic ulcer was 55.5, 69.4 and 84.6%, respectively. There was no significant correlation between the presence of these genes and the type of H.pylori-related diseases.
Conclusion: Frequency of BabA2 and Hpa genes is higher in the samples of peptic ulcer but there was no significant relationship between these genes and H.pylori-related diseases.

Abstract
ABSTRACT
Background and Objectives: Breast cancer is one of the most common cancers among women. Cardiovascular disease is one of the most important complications of breast cancer. The aim of this study was to investigate the effects of six weeks of swimming on malondialdehyde (MDA) and superoxide dismutase (SOD) levels in the heart tissue of mice with breast cancer.
Method: Breast cancer was induced in 30 female Balb/c mice (6-8 weeks old, weighing 150-200 g) by tumor transplantation. The tumor site was examined every day for the formation or absence of tumor. After becoming familiar with the laboratory environment and swimming pool, the subjects were randomly divided into three equal groups: 1. healthy control, 2. cancer control, and 3. swimming-cancer. The swimming group performed swimming training 60 minutes/day, five days a week, for six weeks. After the intervention, the levels of SOD and MDA in the heart tissue samples were evaluated by enzyme-linked immunosorbent assay. Data were analyzed in SPSS (version 16) using independent t-test at significance of 0.05
.
Results: The results showed that induction of breast cancer was accompanied by a significant increase in MDA and a significant decrease in SOD in the heart tissue of mice. Participation in the six-week swimming significantly reversed these effects
.
Conclusion: Swimming could have beneficial effects in improving oxidative factors in breast cancer patients.

Abstract
ABSTRACT
Background and Objectives: Diabetes is one of the most common diseases and a major risk factor for cardiovascular disease. Studies have shown that regular exercise can affect apoptosis in cardiomyocytes. The purpose of this study was to investigate the effect of high-intensity interval training on the expression of Bcl-2 and Bax as important apoptosis factors in diabetic rats.
Methods: After inducing diabetes in 20 male Wistar rats (weighing 250 ± 1.15 g), the rats were randomly divided into a control group and a training group. The training group performed high-intensity interval training five days a week for four weeks, and the control group did not perform any training. After the intervention, RNA was extracted and TCF mRNA was subjected to real time RT-PCR for measuring Bax and Bcl-2 expression in the heart tissue of diabetic rats. Data were analyzed by IBM SPSS Statistics V22 using independent t-test. P-values less than 0.05 were considered as statistically significant.
Results: The expression of Bcl-2 increased significantly and the expression of Bax decreased significantly after the four-week training intervention.
Conclusion: The high-intensity interval training can have beneficial effects on the expression of apoptotic genes in rats with type 2 diabetes.

Abstract
Background and objectives: The prevalence of obesity and overweight in women is increasing. Secretion of adipokines such as vaspin may affect insulin sensitivity. It has been suggested that physical activity can alter circulating vaspin levels. The purpose of this study was to investigate effects of six weeks of aerobic interval training on serum vaspin and blood glucose levels in obese inactive girls.
Methods: In this quasi-experimental study, 22 inactive overweight girls were selected from the Shahid Chamran University of Ahvaz, Iran. The mean age, weight and body mass index of the subjects were 24.22±1.35 years, 83.77±12.66 kg and 32.56±3.36 kg/m2, respectively. The subjects were randomly divided into an experimental group )n=11) and a control group )n=11). The experimental group performed aerobic training on stationary bicycle at 65-80% of maximal strength, three sessions a week for six weeks. Serum levels of vaspin and glucose were measured using commercial kits. Data were analyzed using SPSS (version 23) and at significance of 0.05.
Results: The training intervention had no significant impact on the serum vaspin and glucose concentrations (P>0.05). In addition, the covariance analysis showed that serum vaspin and glucose levels did not differ significantly between the study groups (P>0.05).
Conclusion: Based on the results, the six-week aerobic interval training had no significant effect on vaspin and blood glucose levels of inactive obese girls. It is recommended to further examine the effect of this type of exercise at higher intensities and for longer durations.

Abstract
Background and objectives: As a chronic inflammatory airway disease, asthma can increase level of markers of inflammation and muscle damage. In this study, we aimed to evaluate the effects of short-term continuous aerobic exercise combined with vitamin D supplementation on high-sensitivity C-reactive protein (hs-CRP), lactate dehydrogenase (LDH) and creatine kinase (CK) levels in asthmatic women.
Methods: In this study, 30 overweight (body mass index=26.97± 1.24 kg/m2) women with mild to moderate asthma were purposefully selected.. The subjects received a vitamin D tablet with a daily dose of 1000 IU for six weeks. The training protocol consisted of two sessions of Balke treadmill test (one session before and one session after the vitamin D supplementation). Blood samples were taken before and immediately after the exercise and before and after vitamin D supplementation. Repeated measures ANOVA was used to evaluate changes in the levels of hs-CRP, CK and LDH.
Results: The variables were not significantly affected by the exercise and vitamin D supplementation (P>0.05).
Conclusion: It seems that a single session of short-term aerobic exercise and six weeks of vitamin D supplementation do not significantly affect hs-CRP, CK and LDH levels in asthmatic women.

Abstract
Background and objectives: Fanconi anemia (FA) is an autosomal recessive disorder that usually manifest in forms of pancytopenia, hyperpigmentation, and skeletal complications. Mutation in the DNA repair regulatory genes is associated with the development of FA. Examination of chromosomal breakages when chromosomes are exposed to cross-linking agents is a common method of FA diagnosis. This study aimed to evaluate the prevalence and characteristics of patients with FA in Mashhad, northeast of Iran.
Methods: In this study, we evaluated 312 suspected FA patients who had been referred to the laboratory of Ghaem Hospital during 2014-2020. The mitomycin C method was used to identify FA-positive subjects.
Results: After the examinations, 84 patients (26.9%) were cytogenetically positive for FA. Of 84 patients, 48 (57.1%) were male and 36 (42.9%) were female. Thumb abnormality was the most common congenital anomaly (43.2%).
Conclusion: Based on the findings, males are more susceptible to FA, and thumb abnormality is the most common congenital anomaly associated with FA. Combination of clinical manifestations and genetic susceptibility in patients may contribute to a more accurate diagnosis.

Abstract
Background: Omentin-1 plays an important role in insulin function. Despite numerous studies, the effect of interval training on this adipokine is still vague. This study aimed to elucidate the effect of high-intensity interval training (HIIT) on serum glucose, insulin, insulin resistance (IR), omentin-1 serum levels, and gene expression in the visceral adipose tissue in type 2 diabetic (T2D) rats.
Methods: In an experimental study, 20 male rats (8-10 weeks, weight: 250-270 g) were randomly divided into 2 groups: diabetic control (N=10) and diabetic training (N=10). The training protocol was 30 minutes of HIIT (1-min run, 2-min rest) performed 5 days a week for 4 weeks. Fasting blood glucose, insulin resistance, omentin-1 serum level, and gene expression were measured in the visceral fat 48 hours after the last exercise for both groups.
Results: The HIIT resulted in lower serum glucose and insulin resistance (P = 0.001), higher serum omentin-1 levels (P = 0.001), and higher visceral fat gene expression (P = 0.004) in the training group compared to the control group.
Conclusion: Lower serum glucose and insulin resistance and higher omentin-1 serum levels and gene expression in the training group can prove the effectiveness of HIIT training in T2D, although further research is required.
