Abstract

Abstract Background and objectives: Familial hypercholesterolemia (FH) is an autosomal disorder characterized by increased levels of total cholesterol and low density lipoprotein cholesterol. The FH clinical phenotype has been associated with increased risk of coronary heart disease and premature death. The mutation in LDLR gene in most cases is responsible for FH phenotype. Furthermore, other gene mutations such as apolipoprotein B- gene may cause similar results. Preliminary research indicates that the FH phenotype is also influenced by other genetic and environmental Factors therefore, routine clinical analysis such as total cholesterol and LDL-C levels in serum, for early diagnosis and treatment, are not sufficient. Molecular diagnostic investigations, because of high specifity and sensitivity near %100, administered for determining the prevalent mutations in LDLR (and probably other genes) are needed for exact diagnosis and accurate therapy. Currently, PCR-SSCP and southern blotting techniques are among the common techniques that could detect major mutations in gene. Because of wide diversity in kinds of mutations in LDLR gene, we recommend, first, determining the proband's mutation and kinds of mutation, then, performing routine test based on type of mutation. Abstract

Abstract

         Background and Objectives: Concentration low-density lipoprotein (LDL) is one of the strongest indicators of atherosclerosis and predicts the diagnosis of cardiovascular diseases. LDL measurement accuracy is very important. LDL can be measured directly, such as enzymatic and nephelometry methods or can be calculated using Friedewald's formula. Despite the development of enzymatic methods and LDL nephelometry still in most laboratories is calculated using Friedewald's formula. The aim of this study was an investigation of correlation coefficient between two methods of measuring LDL- cholesterol levels.

         Methods: This descriptive cross-sectional study, performed on the 1141 patients. Cholesterol, triglycerides, HDL, LDL all patients assayed by enzymatic method. For patients with triglyceride levels of less than 400 mg/dl had LDL levels were calculated by Friedewald's formula. Normal levels of LDL/HDL ratio of less than 3.5 were considered.

          Results: Of the 1141 patients participating in this study, 38.3 % men and 61.7 % women. The mean patient age was 46.3 ± 16.1 years. Mean serum cholesterol, triglycerides and HDL were 177.9 ± 41.1, 132.9 ± 73.2 and 45.8 ± 13.2 mg/dl, respectively. Average direct and calculated LDL concentration was 82.1 ± 23.1 and 105.5 ± 35.8, respectively. The direct measurement of LDL, LDL/HDL levels in 97.1% of cases was normal, while 85.1 % of the calculation of LDL were normal. Pearson correlation coefficients were obtained by two methods: 0.869 (p <0.001).

         Conclusion: Despite the favorable correlation between two methods of measurements of LDL, the results of a calculation method is more than direct method. This can have a negative impact on the judgment of the treating physician.

Abstract

ABSTRACT
        Background and Objectives: The unfavorable lipid profile in obese individuals is associated with high incidence of various diseases including cardiovascular disease, hypertension, etc. Dieting for weight loss and physical activity are among the most important factors affecting the serum lipid profile. The aim of this study was to compare the effect of diet with and without physical activity on body mass index and serum lipid profile of obese women.
      Methods: This clinical trial was performed in 2011 on 39 obese women referred to a weight loss and nutrition counseling center. Subjects were randomly divided into an active group (diet and exercise) and inactive group (diet without exercise). Blood sampling was done before the intervention and two months after the intervention. Data was analyzed using SPSS-16 and t-test.
       Results: Mean level of triglyceride reduced significantly in both groups. Mean level of cholesterol and low-density lipoprotein decreased significantly only in the active group. The Mean level of high-density lipoprotein in the two groups had no significant difference.
Conclusion: The results indicate that dieting for weight loss along with short-term physical activity improves serum levels of cholesterol and low-density lipoprotein, but does not affect serum HDL level.

Abstract

Background and objectives: Sedentary life style and high-fat diet are one of the key causes of coronary artery disease. The aim of this study was to evaluate effects of periodic exercise and consumption of fenugreek seed extract on the expression of oxidized low-density lipoprotein (oxLDL) and reactive oxygen species (ROS) genes in patients with coronary artery occlusion.
Methods: The present quasi-experimental study was performed on 32 men (60±5 years) with coronary artery disease. The subjects were randomly divided into four groups: control, exercise, fenugreek supplementation, and combination of exercise and fenugreek supplementation. The training program consisted of eight weeks of intermittent running at 55 to 65% heart rate reserve, three sessions per week, with an emphasis on gradual overload. The subjects consumed 10 mg/kg of body weight fenugreek extract daily. Data analysis was performed using two-way analysis of variance and Tukey's test at statistical significance of 0.05.
Results: The mean expression ratio of oxLDL and ROS genes in the intervention groups decreased significantly (p<0.0001). Compared with the control group, the most profound decreased in oxLDL and ROS expression occurred after combination of exercise and fenugreek supplementation (p<0.0001).
Conclusion: The results showed that intermittent exercise and consumption of fenugreek seed extract might have beneficial effects on the antioxidant defense system in patients with coronary artery occlusion.

Background: Multiple sclerosis (MS) is a demyelination disorder of the central nervous system (CNS), which is believed to be associated with oxidative stress. Therefore, researchers try to find reliable biomarkers to monitor the disease and predict its prognosis. Cholesterol and lipids in the myelin sheath are vital for nerve cells. Serum low-density lipoprotein (LDL) is susceptible to lipid peroxidation induced by oxidative stress. This study aimed to evaluate oxidative stress markers in the serum of patients with relapsing-remitting MS (RRMS) and examine their correlation with lipid markers.
Methods: A total of 18 MS patients (14 women and 4 men) and 18 healthy subjects (matched by age and sex) were enrolled in this cross-sectional study. The serum samples were collected in both relapsing and remitting phases. The prooxidant-antioxidant balance (PAB), malondialdehyde (MDA), and oxidized LDL (oxLDL) were measured as markers of oxidative stress.
Results: The mean age of participants was 29.21 (22-42) years. In the comparison between the patient and control groups, the most differences were increased levels of PAB in the patient group (P < 0.05), no difference between relapsing and remitting phases (P = 0.995), increased MDA levels in the relapsing phase (P = 0.013)––but no change in the remitting phase (P = 0.068), no difference in LDL and oxLDL levels in the patient group (P > 0.05), and MDA, LDL, and oxLDL levels did not have any significant correlation with PAB (P > 0.05).
Conclusion: High levels of oxidative stress markers were present in both phases of the disease. Lipid peroxidation markers (such as MDA) increased in the acute phase, but oxLDL did not change. Also, there was no significant correlation between oxidative stress and cholesterol markers.
